mRNA Synthesis (ECL โ€” Day 1)

In vitro transcription of GFP mRNA with 5' Cap and poly-A tail. Pseudouridine substitution for stability. Expected yield: 50-100 ฮผg.

Lipid Nanoparticle Encapsulation (ECL โ€” Day 1-2)

Encapsulate mRNA in LNPs (ionizable lipid:DSPC:cholesterol:PEG-lipid 50:10:38.5:1.5). Size target: 80-120 nm. Encapsulation efficiency >80%.

Agroinfiltration (Day 3)

Needleless syringe infiltration into abaxial leaf surface of 4-6 week old N. benthamiana plants. Concentration: 0.1-1 ฮผg/ฮผL LNP-mRNA solution.

Observation (Day 5-7)

UV light (365 nm) illumination of infiltrated leaves. GFP fluorescence visible as bright green signal. Photograph with UV filter.

Results & Report (Day 7-10)

Quantify fluorescence intensity. Generate PDF report with images and data. Upload to ParisiVaxAI dashboard.
